The referee has mentioned that it is important to take into account the spectral response of the system. The fluorophores emission, and in particular, the fluorescence curve of NADH is then reproducible. In order to take into account this effect, the experimental curves were corrected by the PMT response. This issue is addressed in the ... section of the manuscript. Figures 3 to 6 have also been redrawn with this correction.


The dominant departure from a flat response in our system came from the photo multiplier tube. The efficiency curve for the ___ model was obtained from the manufacturer. The spectra of NADH, both in PBS solution and in cells, taken with this PMT were divided by the PMT efficiency function in order to produce a flat frequency response.

The PMT becomes rather inefficient towards the red and the corrected curves therefore show a considerable increase in the red part of the spectrum. The uncorrected spectrum of free NADH in PBS solution is shown in figure ___ . In contrast, the corrected spectrum is shown in figure ___+1. The peak wavelength in the latter curve is ___ and the FWHM is ___ consistent with other results [see Croce, etc.]. The corrected spectra was fitted with two Gaussians.
